Nicergoline is an ergot derivative used to treat senile dementia and other disorders with vascular origins.Target: Alpha-1A adrenergic receptorNicergoline acts by inhibiting the postsynaptic alpha(1)-adrenoceptors on vascular smooth muscle. This inhibits the vasoconstrictor effect of circulating and locally released catecholamines (epinephrin and norepinephrin), resulting in peripheral vasodilation.Nicergoline is a semi-synthetic ergot alkaloid derivative. Since its successful development in the 1960s, it has been registered and used in more than 50 countries. It has also been widely used in various diseases such as cerebrovascular disease, dementia and dizziness in China.

Improve cerebral circulation: Nicergoline can dilate resistance blood vessels, thereby improving cerebral circulation. As a specific adrenergic α1 receptor blocker, the drug can inhibit the pressor reflex of resistance vessels, increase the local cerebral blood flow and glucose utilization during the reperfusion period of bilateral internal carotid artery ligation in rats; reduce the blood pressure in anesthetized dogs blood pressure, but does not affect cerebral blood flow; and can improve cerebral vasospasm induced by experimental subarachnoid hemorrhage in rabbits. Improve brain metabolism and electrophysiological activities: Long-term treatment with nicergoline can improve the glucose utilization rate of brain extracts from aged rats. Acute medication can improve memory function in elderly subjects, increase the total energy of EEG topography, and reduce slow waves.
