Rupatadine Fumarate (FR, RupatadineFumarate) chemical name is 8-chloro-6,11-dihydro-11-[1-[(5-methyl-3-pyridine)methyl]-4-piperidine yl]-5H-benzocycloheptano[1,2-b]pyridine fumarate. It was successfully developed by the Spanish pharmaceutical company Uriach and was first marketed in Spain in 2003. It is a new generation of tricyclic antihistamines with dual effects of antihistamine and antagonism of platelet activating factor (PAF). It is also the only powerful and highly effective anti-allergic drug that has both antihistamine effect and PAF antagonistic activity currently on the market.

Rupatadine fumarate is a selective histamine H1 receptor inverse agonist and PAF antagonist. In animal experiments in mice, rats, guinea pigs, rabbits and dogs, the drug showed strong antihistamine effects and PAF receptor blocking effects. The ileal function test of guinea pigs was used to compare the activity of rupatadine and other antihistamines, and the results showed that rupatadine was more active than terfenadine, loratadine, cetirizine, hydroxyzine, Diphenhydramine is stronger and comparable to desloratadine. In a single-dose expansion trial in healthy people, 10 mg of rupatadine inhibited PAF-induced erythema with a mean maximal inhibition of 41% and 20 mg of 56%. In addition, this product has inhibitory effects on mast cell degranulation, neutrophil and eosinophil migration, and cytokine release.
