Riluzole has the following physiological functions: 1. Regulation of glutamate Riluzole is an important glutamate regulator, which affects the concentration of glutamate in the intercellular space from different links. It affects the concentration of glutamate by affecting the activity of the glutamate transporter. It can attenuate the activity of excitatory amino acid transporter 3, and has a linear relationship in the concentration range of 0.3-100 μmol·L-1 (the concentration given to oocytes), indicating that riluzole inhibits the activity of protein kinase C (PKC). to attenuate the activity of excitatory amino acid transporter 3. 2. Neuroprotective effect In a variety of animal models of neuronal damage mediated by excitotoxicity, riluzole showed damage repair function, and it could also promote the production of various nerve growth factors. In a spinal muscular atrophy model, it ameliorated the defective neuroprotective behavior in a spinal muscular atrophy model through small-conductance calcium-activated potassium channels. 3. Effects on nerve discharge Riluzole reduces the repetitive discharges induced by continuous current stimulation at different concentrations, but has less effect on the discharges induced by transient currents. Riluzole can activate TRPC5 heterologous expression in HEK293 cells and endogenous expression in U-87 glioblastoma cell line. Although riluzole activates other ion channels, it does not activate other TRPC members. Thus, riluzole is a useful pharmacological tool to specifically identify TRPC5 currents in immortalized cells and acutely isolated primary cells. 4. Antidepressant studies have found that riluzole combined with antidepressants can significantly improve the condition of patients with refractory depression. The linear mixed model shows that there is a certain relationship between treatment time and treatment effect. After 1 week of treatment, the effect is significant, And it lasts for 12 weeks. In the treatment of bipolar depression, riluzole combined with lithium has achieved significant effect, and the treatment process did not cause the occurrence of mania. 5. Anti-anxiety Numerous evidences confirm the important role of glutamatergic neurotransmitter system in the treatment of mood and anxiety-like diseases. Riluzole can regulate the concentration of glutamate in different links, so it is used for anti-anxiety research. To study the relationship between hippocampal volume and hippocampal N-acetylaspartate during the treatment of generalized anxiety disorder with riluzole; preliminary findings suggest that hippocampal N-acetylaspartate and hippocampal volume are related to glutamate drug treatment response related neural biomarkers. 6. Three Na+ channel blockers, including bisphenylhydantoin, riluzole, and lidocaine, reduce the epileptiform activity of the hippocampus through different mechanisms of action. The three compounds act on different voltage-gated sodium channel sites, but produce similar reductions in reticular dominant excitability. Diphenylhydantoin and riluzole continued to exert their effects on network activity changes until the drug completely disappeared, but lidocaine increased hippocampal CA3 inter-burst intervals (IBIs) only at higher concentrations ). Therefore, their function does not permanently lead to the return of hippocampal CA3 bursts to zero. These data reveal that hippocampal CA3 bursts provide a relatively simple and rapid platform to identify compounds that attenuate network excitability, providing an initial screen for further more complex in vivo animal studies. 7. Analgesic effect Dorsal root ganglion (DRG), as the first-order neuron of pain afferents, plays an important role in the peripheral mechanism of pain perception. Studies have shown that riluzole can promote the growth of DRG neurites.
