Pitavastatin calcium is the first fully synthetic HMG-CoA reductase inhibitor jointly developed by Nissan Chemical and Xinghe Co., Ltd. Inhibition of liver enzymes to reduce the ability of the liver to produce cholesterol, thereby improving the elevated blood cholesterol level, is mainly used for the treatment of patients with hypercholesterolemia and familial hypercholesterolemia, and its lipid-lowering effect is very good. The most powerful lipid-lowering drug so far.

Inhibition of HMG-CoA reductase: Pitavastatin calcium has a strong antagonistic inhibitory effect on HMG-CoA enzyme, the IC50 value is 6.8nmol/L, and its effect is 24 times that of simvastatin and 68 times that of fluvastatin. Block cholesterol synthesis: It can effectively inhibit the process of cholesterol generation in human hepatocytes HepG2, with an IC50 value of 5.8nmol/L, and its effect is 29 times stronger than simvastatin and 57 times stronger than atorvastatin. However, the inhibitory effect of pitavastatin calcium on the enzymes in the process of cholesterol production after mevalonate production is very weak. Increase the density of LDL receptors: Pitavastatin can induce the synthesis of LDS receptor mRNA at an ultra-low concentration of 1 μmol/L, increase the number, and increase the density of LDL receptors, thereby promoting the clearance of LDL and making plasma LDL. - Decreased cholesterol concentration and plasma total triglyceride concentration.
