As a third-generation EGFR inhibitor, osimertinib is aimed at the subgroup of patients with EGFR T790M mutation to meet such unmet market needs: osimertinib has high selectivity, and is highly selective for T790M/L858R mutation and exon 19 deletion of EGFR. The IC50s were 11.4 nM and 12.9 nM, respectively, while the inhibitory activity against wild-type EGFR (IC50) was 494 nM. Osimertinib is an irreversible inhibitor of EGFR and can form covalent bonds with molecular targets, so theoretically the response lasts longer and the chance of drug resistance is reduced accordingly.
